چکیده

A simple, 30-mm method of estimating the binding of bilirubin to serum protein was compared with a quantitative method. Good correlation was found between the two methods, both based on Sephadex gel filtration. The results indicate that the presence of loosely bound bilirubin can be determined with this simple, qualitative, semimicro-scale test. This determination is important for the clinical evaluation of jaundiced infants, since it may help to determine whether and when prophylactic therapy to prevent bilirubin encephalopathy is appropriate.

The influence of clinical status on total bilirubin binding capacity in newborn infants.

The relationship between total bilirubin binding capacity (TBBC) and clinical status was investigated in order to assess the risk of bilirubin toxicity in 83 infants with jaundice in this study. Infants with respiratory distress, acidosis, hypoglycaemia, sepsis, asphyxia-anoxia and hypercarbia were accepted as ill and the remainders were well. Sephadex G-25 gel filtration method was used to det...

Correlation between arginyl residue modification and benzodiazepine binding to human serum albumin.

Human serum albumin (HSA) has been chemically modified with 1,2-cyclohexanedione and N-acetylimidazole under nondenaturing conditions. Derivatives, in which 10 arginine residues (1,2-dihydroxycyclohex-1,2-ylene (DHCH)-HSA), 56 to 57 lysine and 5 tyrosine residues (acetyl-HSA), or 56 to 57 lysine residues alone (O-deacetyl-HSA) are modified, have been isolated. Their conformation has been tested...

Faster estimation of reserve bilirubin binding capacity of serum from the neonate by thin-layer chromatography on sephadex.

Knowledge of the reserve bilirubin-binding capacity is useful in management of hyperbilirubinemia in the newborn. We describe thin-layer gel chromatography that permits measurement of this variable, with a 250-mul sample of serum, within 50 min. Single samples from four different infants can be analyzed on the same plate. The capacity of 40 sera from jaundiced and of 19 sera from nonjaundiced i...

Methods for studying the binding of aluminum by serum protein.

We describe methods for studying the binding of Al by protein in serum: ultrafiltration, gel filtration, and immuno-affinity chromatography. For ultrafiltration we used an Amicon YM10 cellophane membrane with a nominal cutoff of 10 000 Da to separate ultrafiltrable and non-ultrafiltrable Al. For gel filtration we used Sephacryl S-300, and for immuno-affinity chromatography we used anti-transfer...

Comparison of two gel filtration chromatography resins for the purification of foot-and-mouth disease virus as a purified vaccine antigen

Introduction: Foot-and-mouth disease (FMD) is a highly contagious and economically devastating viral disease of livestock that is categorized in list A of animal diseases by the World Organisation for Animal Health (OIE). Vaccination is effective against FMD and the vaccine production centers largely use the industrial ultra-filtration and chromatography in order to remove the cellular proteins...
